Cell and tissue transplantation is a new, rapidly developing research field aiming at regenerate impaired myocardium when treating cardiovascular diseases. To date, there are no accurate and efficient methodologies for cardiomyocytes differentiation, which is needed for identifying safe and strict quality control conditions in practicing stem-cell therapy. When developing new methodologies, it is important to know whether the cell lines, which grow under the same conditions, develop distinctive properties during the differentiation. The differentiation can be followed by studying two most important indicators of the differentiation - morphological and energy metabolism changes in the differentiating cells.
The aim of this work is to compare mesenchymal stem cell lines during their differentiation, through evaluation of morphological and energy metabolism changes in the differentiating stem cells.
The cells were derived from skeletal muscles of three rabbits and were cultivated into three myogenic stem cell lines (FR3, D7, D12) When the differentiation was induced, the morphological changes and NAD(P)H fluorescence were observed, and cell resistance to stressors (60° C temperature) was measured. The obtained results showed that cell populations were very heterogeneous, different in size and differentiation rate (the dynamics of the cell size change in population), and the degree of the changes in population heterogeneity. Increase in fraction of larger size cells during the differentiation was characteristic for all cell lines. The size of cells in the populations grew to the certain size, which was followed by the tendency of the size decrease. During the differentiation, in all tested myoblast lines NAD(P)H fluorescence increased significantly. The differences in sensitivity of non-differentiated myoblast lines to stressors (60° C temperature) were insignificant, but the sensitivity and differences between the lines increased with the differentiation.
